Growth inhibition of purine requiring mutants of Escherichia coli by 5-hydroxyuridine.

چکیده
Substituted pyrimidine nucleosides (1) inhibit the biosynthesis of nucleic acid in several biological systems. Rafelson et al. (2) observed that chlorouridine inhibits the uptake of P3’04 by the ribonucleic acid fraction of minced, 1 day-old, mouse brain. Visser et al. (3) found that a number of substituted nucleosides including amino-, chloro-, diazo-, formamido-, methyl-, and hydroxyuridine inhibit the propagation of Theiler’s GD VII strain of murine encephalomyelitis virus in tissue culture of 1 day-old mouse brain. Fukuhara and Visser (4) and Roberts and Visser (5) reported that several substituted nucleosides inhibit the utilization of uridine, cytidine, or uracil in Neurospora crassa 1298. Recent studies by Werkheiser and coworkers* reveal that Ps204 uptake into various acid insoluble fractions of rat hepatoma is markedly inhibited by aminouridine. The present paper reports the further use of selected nucleoside antimetabolites as a means for studying the interrelations of nucleic acid precursors in various microorganisms with purine and pyrimidine requirements.

Degradation of substituted pyrimidine nucleosides by Escherichia coli and Bacillus subtilis.
It has been shown previously that hydroxyuridine competitively inhibits the growth of purine-requiring mutants of Escherichia coli (1). However, the inhibition is ultimately overcome, presumably because of the destruction of hydroxyuridine. It was of interest to det.ermine whether the inhibitor is, indeed, degraded, and, if so, what structural changes produce the loss of inhibitory activity. In...

متن کاملStudies on the accumulation of 4-amino-5-imidazole carboxamide in Escherichia coli.
When cultures of Escherichia coli are partially inhibited by sulfonamides they accumulate a diazotizable and nonacetylable amine in the medium (Fox, 1942). The amine was isolated by Stetten and Fox (1945) and characterized by Shive et al. (1947) as 4-amino-5-imidazole carboxamide. The latter authors suggested that the amine was a precursor of purines as it only required the addition of a one ca...

متن کاملExpression of tolC and organic solvent tolerance of Escherichia coli ciprofloxacin resistant mutants
AcrAB-TolC is a major efflux pump in Escherichia coli. It was reported that tolC is overexpressed and involves in improving the organic solvent tolerance level in Escherichia coli marR mutants that are resistant to several antibiotics, such as ciprofloxacin. Low and intermediate levels resistance did not improve organic solvent tolerance. Thus, in this descriptive-experimental study it was deci...
